RESUMO:

Bovine brucellosis is a bacterial zoonosis that affects livestock and public health and in Brazil it is caused by Brucella abortus. The disease causes reproductive clinical signs in cattle such as abortions and premature births and nonspecific clinical signs in humans such as fever, arthralgia, night sweats, among others. The infection is not treated in cattle, and the slaughter of positive animals is recommended. In humans, treatment is difficult, and the combination of synergistic antimicrobials for periods of 4 to 6 weeks is proposed. The aims of these works were (i) to describe the identification and complete sequencing of the first strain of Pseudochrobactrum sp. genus isolated in Brazil, previously classified as B. abortus; (ii) identify the genetic mechanisms associated with the antimicrobial resistance and multidrug resistance phenotype in B. abortus strains naturally resistant or multidrug resistant to drugs commonly used for the treatment of human brucellosis (iii) perform pangenome analysis of 53 samples of B. abortus isolated in Brazil that had their complete genome sequenced and 21 complete genomes of this species deposited on the NCBI platform. As preliminary results we obtained the description of the main characteristics of the genome of Pseudochrobactrum sp. strain, as well as the construction of a phylogenetic tree relating the sample to other representatives of the Brucellaceae family. In addition, the results and analysis strategies for genetic determinants of antimicrobial resistance have been described in detail, as well as their possible limitations for organisms belonging to the genus Brucella. Finally, pangenome analysis made it possible to identify that the 74 representative genomes of B. abortus are highly conserved, based on the observation of the core genome and the essential genes of the species
